Made famous in the film lock, stock and two smoking barrels, this phrase has been around for a long time.
It means to put everything into something, so if you have got your head down and are really focussing on your target, then you can be said to be doing something lock, stock and barrel.
Now the phrase comes from the world of guns and weapons. Rifles have the three compartments mentioned, and you need all three for them to be useful, in isolation they have no purpose.
Therefore with wholeness and combining the various necessary elements you were said to be doing something lock, stock and barrel.
